Ebersole, who also nabbed a Tony for her 2001 turn in "42nd Street," will play Elvira, the titular spirit who haunts her first husband in the 1941 Noel Coward comedy.
No further casting has been announced, although Angela Lansbury and Rupert Everett are said to be on the wish list. Michael Blakemore ("Noises Off," "Copenhagen"), who recently helmed Lansbury in "Deuce," will direct.
"Spirit" begins perfs Feb. 26 ahead of a March opening at a Shubert theater to be determined.
Revival is produced by Jeffrey Richards, Jerry Frankel and Steve Traxler, the team also behind the revival of David Mamet's "Speed-the-Plow" that begins previews Oct. 3.
