"Along Came Polly" helmer John Hamburg, who inked a two-year overall deal with DreamWorks in April, will direct.
Hamburg-penned script centers on a man about to get married who actively seeks out a male friend to be his best man for the wedding.
Shooting is scheduled to start in March.
Donald De Line is producing alongside Hamburg. Ivan Reitman, Tom Pollock, Bill Johnson and Jeff Clifford exec produce.
Rudd and Segel last teamed up in Judd Apatow's summer comedy "Knocked Up." The pair also recently wrapped another Apatow project, "Forgetting Sarah Marshall," which Segel penned.
Rudd next appears in the Apatow-produced John C. Reilly starrer "Walk Hard: The Dewey Cox Story." His credits also include "The Ten," which he produced, and the upcoming "Little Big Men," which he co-wrote.
Segel is a star on CBS' "How I Met Your Mother."
