Movie centers on the life of Klaus Barbie, the Nazi and "Butcher of Lyons" who lived undisturbed in Bolivia for many years after World War II, but was caught, tried and convicted in the 1980s for war crimes.
Doc is said to focus particularly on Barbie's involvement with the CIA, which reportedly employed him for years.
Marcel Ophuls' earlier doc about Barbie, "Hotel Terminus," won an Oscar in 1989.
TWC has bought all U.S. and Canadian rights; no release date has been set.
Movie marks a return for Macdonald to the documentary form. After directing a number of acclaimed films, including the nature doc "Touching the Void" and Oscar winner "One Day in September," Macdonald moved into features with "The Last King of Scotland."
