Studio is keeping the plot under wraps.
Thriller marks the first film developed under Flower Films' new producing pact with Warners. Shingle, based on the studio's Burbank lot, inked the first-look deal in April.
Producers of "He Loves Me" are Barrymore and Nancy Juvonen, her partner at Flower Films.
Warner Bros. Pictures' Kevin McCormick will oversee for the studio.
Project was brought in by Flower Films VP of development Gwenn Stroman.
Flower Films previously had a first-look deal with Sony, producing both "Charlie's Angels" features, "50 First Dates" and "Riding in Cars With Boys." Most recently, the shingle produced "Fever Pitch" for 20th Century Fox.
Barrymore stars opposite Eric Bana in Warners' upcoming gambling pic "Lucky You," directed by Curtis Hanson and written by Eric Roth. She also is voicing one of the roles in Universal's animated pic "Curious George."
