The romantic comedy is a loosely inspired remake of the 1972 pic written by Neil Simon and directed by Elaine May.
Story concerns a man who hastily weds a woman he thinks is perfect -- until he falls in love with another woman during their honeymoon.
Radar Pictures founder Ted Field will produce with Bradley Thomas, the Farrellys' longtime producing partner at Conundrum Entertainment.
John Davis and Radar's Joe Rosenberg will exec produce.
A few helmers have been attached to the project over the past year, including Barry Sonnenfeld and James Bobin. Jason Bateman and Amy Poehler were also recently set to star.
With the Farrellys aboard, however, the film will be packaged anew.
The Farrelly brothers remain committed to directing "The Three Stooges" but haven't yet nailed down how they want to put the picture together.
Most recently, the siblings directed Fox 2000's baseball-themed romantic comedy "Fever Pitch." They're also exec producers on "The Ringer," Fox Searchlight's upcoming comedy starring Johnny Knoxville as a wannabe Special Olympian.
